Killing a target will lock out all other opportunities related to them for that playthrough but those for other targets will remain usable. Most of them will complete before you actually use them to make a kill (Catharsis, for instance, unlocks once you get into Silvio's room). Follow the prompts to create an opportunity. Press the to bring up the menu then go to the Opportunities tab, now select one and it will be highlighted in the mission. You can easily work your way through them by tracking them as you start the level.
